    Numerical study of a slip-link model for polymer melts and nanocomposites

    We present a numerical study of the slip link model introduced by Likhtman for describing the dy- namics of dense polymer melts. After reviewing the technical aspects associated with the implemen- tation of the model, we extend previous work in several directions. The dependence of the relaxation modulus with the slip link density and the slip link stiffness is reported. Then the nonlinear rheolog- ical properties of the model, for a particular set of parameters, are explored. Finally, we introduce excluded volume interactions in a mean field such as manner in order to describe inhomogeneous systems, and we apply this description to a simple nanocomposite model. With this extension, the slip link model appears as a simple and generic model of a polymer melt, that can be used as an alternative to molecular dynamics for coarse grained simulations of complex polymeric systems

    COMPLEXIDADE RACIAL: mitos e realidades em duas freguesias de Salvador em 1775

    A partir da análise minuciosa dos dados do Censo de 1775 sobre duas freguesias de Salvador (São Pedro e Penha), são colocados em questão cinco mitos dominantes sobre a escravidão no imaginário nacional: (1) o domínio total do trabalho escravo na sociedade; (2) uma sociedade formada apenas por senhores e escravos; (3) uma sociedade constituída, por um lado, por um segmento de dominantes e exploradores e, por outro, por dominados e explorados; (4) uma sociedade urbana segregada; (5) uma sociedade patriarcal, em que as mulheres eram submissas e economicamente subordinadas. Os resultados do censo, portanto, levantam novas questões para o entendimento da complexidade do nosso passado, o que ajuda a entender a manutenção das extremas desigualdades atuais, além de evidenciar a existência de diferenciações espaciais na cidade. The results of the census, therefore, bring new subjects to understanding the complexity of our past, what helps to understand the maintenance of the extreme current inequalities, besides showing the existence of space differentiations in the city.     Entanglement-induced reinforcement in polymer nanocomposites

    International audienceWe propose a coarse-grained model able to describe filled entangled polymer melts. Our purpose is to study the reinforcement caused by the effect of fillers on the entanglement network, as speculated in previous experimental work, and also observed in molecular dynamics studies. In this work, the filler volume fraction effect, the distribution of the fillers (cubic lattice, randomly dispersed, and small clusters randomly dispersed) and the presence or absence of grafted chains on the fillers are investigated. Our model is based on a “slip-link” model initially developed to study the entanglements in pure polymer melts and offers a less costly computational method than molecular dynamics simulations for the study of entangled polymer melts. The polymer chains are described as Rouse chains of Brownian particles connected by Hookean springs, and are subject to friction and random forces. Entanglements are artificially imposed by objects (slip-links) exhibiting statistical fluctuations that do not modify the equilibrium statistics of the melt. In addition we introduced excluded volume interactions between chain segments, to take into account the incompressibility of the melt. These excluded volume interactions do not perturb the dynamics of the chains in the homogeneous limit as expected from theoretical considerations on short range interactions. Finally, the fillers are modeled by immobile spherical objects, with or without grafted chains, which interact with a repulsive potential with the chain monomers. The chains grafted onto the fillers are represented by “additional slip links” confined in the vicinity of each filler. We first present the effect of the filler distribution and filler volume fraction, considering only bare fillers. Then, the effect of grafted chains via the additional slip-links is also shown as a function of the same parameters. Our results show that the presence of grafted chains induces an important change in the viscosity, calculated by integrating the stress autocorrelation function. Both the plateau value and the terminal relaxation time depend on the density of fillers and on the number of grafted chains. Moreover, we find that a disordered filler configuration induces confinement effects that amplify reinforcement compared to the case of a perfectly ordered configuration

    Biochemical analysis and immunogenicity of Leishmania major amastigote fractions in cutaneous leishmaniasis

    Soluble Leishmania antigen (SLA) from both developmental stages of L. major (L. major MRHO/IR/75/ ER) were prepared. Three and five subfractions of SLA from amastigote and promastigote were obtained by fast protein liquid chromatography (FPLC), respectively. Biochemical analyses and comparison of amastigote and promastigote SLA were done. The biochemical analyses revealed that the first fraction of L. major amastigote possesses a distinct band on its electrophoretic mobility pattern corresponding to a position of 24 kD, and it has enzymatic activity with characteristics of a cysteine proteinase. The isolated fractions of amastigote were tested for induction of proliferation, interferon-gamma (IFN-γ) and IL-4 production in cultures of peripheral blood mononuclear cells (PBMC) from individuals who had recovered and also chronic patients of cutaneous leishmaniasis caused by L. major. The cells of recovered individuals compared with chronic cases proliferated profoundly in response to the first fraction of amastigote SLA. In all recovered individuals, the IFN-γ, but not IL-4, was secreted in response to stimulation with the first fraction of amastigote SLA. In chronic cutaneous leishmaniasis, IFN-γ was infrequently observed in response to stimulation by all three fractions of amastigote SLA, but secretion of IL-4 was observed. These data indicate that first fraction of amastigote SLA is a strong inducer of primed human immune response to L. major, and may have a protective function

    Platelet and Erythrocyte Sources of S1P Are Redundant for Vascular Development and Homeostasis, but Both Rendered Essential After Plasma S1P Depletion in Anaphylactic Shock

    International audienceRATIONALE:Sphingosine-1-phosphate (S1P) signaling is essential for vascular development and postnatal vascular homeostasis. The relative importance of S1P sources sustaining these processes remains unclear.OBJECTIVE:To address the level of redundancy in bioactive S1P provision to the developing and mature vasculature.METHODS AND RESULTS:S1P production was selectively impaired in mouse platelets, erythrocytes, endothelium, or smooth muscle cells by targeted deletion of genes encoding sphingosine kinases -1 and -2. S1P deficiency impaired aggregation and spreading of washed platelets and profoundly reduced their capacity to promote endothelial barrier function ex vivo. However, and in contrast to recent reports, neither platelets nor any other source of S1P was essential for vascular development, vascular integrity, or hemostasis/thrombosis. Yet rapid and profound depletion of plasma S1P during systemic anaphylaxis rendered both platelet- and erythrocyte-derived S1P essential for survival, with a contribution from blood endothelium observed only in the absence of circulating sources. Recovery was sensitive to aspirin in mice with but not without platelet S1P, suggesting that platelet activation and stimulus-response coupling is needed. S1P deficiency aggravated vasoplegia in this model, arguing a vital role for S1P in maintaining vascular resistance during recovery from circulatory shock. Accordingly, the S1P2 receptor mediated most of the survival benefit of S1P, whereas the endothelial S1P1 receptor was dispensable for survival despite its importance for maintaining vascular integrity.CONCLUSIONS:Although source redundancy normally secures essential S1P signaling in developing and mature blood vessels, profound depletion of plasma S1P renders both erythrocyte and platelet S1P pools necessary for recovery and high basal plasma S1P levels protective during anaphylactic shock
